Niclosamide is a chemical known since 1959 that has been used as an anthelminthic agent for the treatment of cestodosis (diseases caused by tapeworms).
It has a detrimental effect on the broad lancet, dwarf and bovine tapeworms and other cestodes. But at the same time, it does not have a special effect in the fight against other types of worms, including round worms (pinworms, ascaris, etc.). Pharmacokinetics
Niclosamide is effective against intestinal tapeworms that affect humans and animals. It inhibits oxidative phosphorylation in the mitochondria of worms, leading to depletion of energy reserves.
In addition, niclosamide paralyzes the worms during direct contact, and also reduces their resistance to proteolytic enzymes of the gastrointestinal tract. Thanks to the latter, dead worms are partially broken down by the action of gastrointestinal enzymes, and their remains are excreted in feces.
The drug is slightly absorbed from the gastrointestinal tract. The main effect on parasites is in the area of the small and large intestine. Does not undergo metabolism. The half-life of the drug from the human body is 2 to 5 hours.
With teniarinhoses (infection with bovine tapeworm)
For children under 2 years old - 500 mg per day, 5-12 years old - 1 g per day, for children over 12 years old and adults - 2-3 g per day. The duration of admission is determined by a parasitologist depending on the type and severity of the infection.
With diphyllobothriasis (infection with a wide tapeworm)
Adults: 1 g after breakfast and 1 g one hour later.
Children: under 2 years of age - 250 mg after breakfast and 250 mg one hour later; 2-6 years old - 500 mg after breakfast and 500 mg an hour later.
With hymenolepiasis (infection with dwarf tapeworm)
Adults: Initially, 2 g on the first day, followed by 1 g per day for 6 days.
Children: under 2 years old - 500 mg on the first day, followed by 250 mg per day for 6 days; 2-6 years - 1 g on the first day and then 500 mg a day for 6 days.
Admission rules
Taking additional medications (laxatives, other anthelmintics) or taking other measures (diet, fasting, enemas) are not required before, during or immediately after taking niclosamide. The product is taken on an empty stomach (either 1 hour before or 2 hours after a meal) or after a light meal (preferably after breakfast).
The tablets should be chewed or crushed thoroughly and then swallowed with a little water. If niclosamide is given to young children, the tablets are ground into powder and mixed with a little water to form a paste.
Mechanism of action
Niclosamide has the maximum therapeutic effect on tapeworms, especially effective against bovine tapeworm and broad tapeworm. The active ingredient niclosamide prevents the absorption of glucose by parasites, which leads to immobilization of the worms: they stop feeding, slow down their own vital functions, and then leave the body through the intestines along with feces. The latter explains the need to use a laxative together with Niclosamide.
The drug is poorly absorbed in the gastrointestinal tract and excreted through the intestines, partially metabolized. The main effect of the tablets is provided on the small and large intestinal regions. The half-life of the active ingredient from the human body is 2 to 5 hours. Since some people have an intolerance to the substance niclosamide, only a parasitologist is authorized to prescribe the medication.
Indications for use
It is forbidden to carry out treatment with Niclosamide without medical advice, and before prescribing the drug, the specialist performs all the necessary tests to determine the type of helminths.
